If Maria doesn't kill her initial target, can she pick another character as a target?

You can only choose at the beginning of the game and once she destroys her target, so... no?

No she cannot. Otherwise she'll change her target at the start of every turn.

According to my reading of the rule, she can't (and therefore loses the ability, sorta).

Well, the rule clearly states that she have to destroy the target. So yes, if someone else will kill the target, then ability is lost.
